From 1991 to 2000, he was the lead singer of the iconic rock band Rage Against the Machine. In 2008, he founded the band One Day as a Lion with Mars Volta drummer Jon Theodore.

During his early teenage years, he and his boyhood friend and future Rage Against the Machine bassist Tim Commerford played in a local band called Juvenile Expression. De la Rocha afterwards joined an underground band named Inside Out and performed with them.
